Genetic and mutagenicity assessment

Genotoxicity tests are in vitro and in vivo tests used to detect DNA damage in subjects that induce genetic damage directly or indirectly through different mechanisms. Fixation of DNA damage in the form of gene mutations, larger-scale chromosomal damage, or recombination is often considered to underlie heritable effects. Lifeasible will select a reasonable test method based on the structural characteristics, physicochemical properties, and available information from pharmacological and toxicological studies of the subject plant extracts, design an appropriate test protocol, and conduct a comprehensive analysis and evaluation of the test results.

  • Ames test service
    We apply the Ames test method to detect the mutagenic effects of the tested plant extracts with and without the addition of an in vitro metabolic activation system (S9) to evaluate their potential genotoxicity. We provided strains including TA98, TA100, TA1535, TA1537, TA97, TA97a, TA102, WP2 uvrA and WP2 uvrA (pKM101). Items identified include, but are not limited to, the following:
    • Identification of histidine-deficient phenotype (his-)
    • Identification of lipopolysaccharide barrier defective (rfa)
    • Identification of uvrB repair-deficient phenotypes
    • Identification of R-factor (ampicillin-resistant)

  • Bone marrow polychromatic erythrocyte (PCE) micronucleus assay service
    We apply the mammalian (e.g., mouse) erythrocyte micronucleus assay to detect chromosome damage or mitotic apparatus damage induced in mouse erythroblasts by test plant extracts to evaluate their potential genotoxicity. It can be used for biological materials of any karyotype, with high confidence in the test results, can detect abnormal chromosomal effects for a long time, and can detect spindle toxicants. It has the advantages of easy observation, clear specimen background, no need for chemical substances other than the compound being tested, and is economical, simple, and rapid.
  • Bone marrow polychromatic erythrocyte (PCE) micronucleus assay service

  • Mouse sperm abnormality test service
    We help you study the toxic effects of plant extracts on animal sperm cells by observing changes in sperm status in mice exposed to the plant extracts to be tested for a period of time. This is one of the common methods of mutagenesis of germ cells.
    • Sperm density: assesses the number of spermatozoa per unit volume of semen.
    • Sperm viability: assesses the motility and vigor of spermatozoa.
    • Sperm morphology: assesses the morphology and structure of spermatozoa, including the morphology and structure of the head, neck, and tail.
    • Sperm abnormality rate: assesses the proportion of spermatozoa with abnormal morphology and structure.
    • Sperm count: assesses the number of spermatozoa per unit volume of semen.
    • Spermatogenesis: assesses the incidence and maturity of spermatozoa.
  • Other programs to detect genotoxicity and mutagenicity of plant extracts
    DNA adducts, which are electrophilic compounds or their metabolites that form covalent linkages with DNA molecules in living organisms, are the most important and prevalent form of chemical damage to DNA. In addition to the above testing services, we offer a DNA adduct-based technique that can be performed using high-performance liquid chromatography-mass spectrometry (HPLC-MS/MS), which is capable of efficiently screening for genotoxic constituents in the tested plant extracts.
